Media Preparation

Description
Since 1985, IMBB maintains a self-contained Cell Culture Facility that houses all equipment needed for the growth, maintenance, and analysis of animal cells. The facility provides basic support to investigators: preparation, filtration and testing of numerous cell culture media and is a source of culture supplies and sterile reagents. The facility has experience in: a) growing a range of different cell lines and hybridomas including mammalian and insect cell lines as well as primary cells b) large scale cell cultures c) transfection of cells and expansion of different clones d) gene targeting and e) embryonic stem cells. Moreover, laser based methodologies are examined for tissue engineering.

The main facility is located on the 1 st floor (Room A208) of the IMBB building and has three separate rooms with constant HEPA filtered air flow that is decontaminated by UV irradiation daily. A second facility is housed in the basement of the Institute and accommodates insect cell cultures, the parasite transgenesis laboratory and the tissue engineering activities.

The cell culture facility complies with all the EU regulations regarding biohazard material handling and disposal: Most of the consumables used in cell culture are disposable and the non disposable ones are autoclaved regularly. The liquid waste are collected in vacuum bottles containing chlorine and disposed in a special sink. The contaminated sharp items are collected in special plastic boxes and disposed together with the biohazard material.

Equipment
The cell culture facility contains:

•  Ten laminar flow hoods, biosafety level II for the sterile handling of cells,
•  Ten CO2 , 37o C incubators for growing mammalian cells
•  Five 25o C incubators for insect cell lines
•  Four refrigerated centrifuges
•  Three inverted phase-contrast fluorescence microscopes with camera and four standard bright-field microscopes for inspection of cell cultures and imaging.
•  Two electroporators
•  Two water baths
•  A highly purified water supply, a conductivity meter and a pHmeter for media preparation
•  A cold room and a freezer for storage.
•  Three liquid nitrogen tanks (with a capacity of more than 5000 ampoules) for storage of cell stocks and clones.
•  A flow cytometer

The Facility also incorporates a dedicated service room, which houses a bottle-washer, sterilizing oven and autoclave.

Services
The facility can provide researchers with:
•  Technical advice and service for culturing a range of cell types
•  Frozen stocks of cells
•  Media preparation, filtration and testing
•  Training in cell culture techniques including cell transfection
•  Mycoplasma screening
•  Microbial contamination testing
•  Monoclonal antibodies from specific hybridoma cells

Facility staff can provide instruction in basic aspects of animal cell culture and aseptic technique. Specialized training is also available for techniques covering the full range of cell culture, including:

•  Cell preservation
•  Mycoplasma detection
•  Cell cloning
•  Hybridoma production
•  DNA transfection
•  Toxicity testing
•  Mass cell culture using microcarriers, suspension, roller or hollow fiber procedures

Guidelines for Facility Equipment Usage
•  All prospective clients must schedule in advance any visit to a Facility or usage of its equipment. The Facility Scientist in charge may waive this requirement depending upon circumstances.
•  All first time clients must register with the appropriate Facility Scientist for training in the usage of equipment assigned to a Facility. The Facility Scientist may waive this requirement if the client can demonstrate to the Scientist's satisfaction adequate knowledge concerning equipment usage and related safety protocols.
•  Clients will be charged fees for damages or losses resulting from any unauthorized or inappropriate usage of a Facility's equipment, supplies, or support systems. However, the Facility Scientist at his or her discretion may waive charges, including regular usage fees, for malfunctions or other failures that are unrelated to the client's use of the equipment.
•  No client should make changes or other alterations to a Facility's equipment without the prior approval and/or supervision of the appropriate Facility Scientist.
•  Clients who need to cancel a scheduled appointment should notify the appropriate Facility Scientist as soon as possible. At the discretion of the Facility Scientist, a client's inability to meet a scheduled appointment may result in financial penalties. Repeated failures to meet scheduled appointments may also result in the loss of privileges to access the equipment.
•  Clients that are more than 15 minutes late for an appointment may lose their scheduled slot to the next individual scheduled to use the instrument, and will need to reschedule their appointment.
•  Unless indicated otherwise by the Facility Scientist, clients will clean work areas and remove any unwanted materials upon completion of use of equipment in the Facility.
•  Clients will notify immediately the Facility Scientist of any equipment malfunctions or other unusual equipment activity (see posted emergency contact information).
