Εξετάζουμε τους μηχανισμούς ρύθμισης γονιδίων που αποκρίνονται σε περβαλλοντικές αλλαγές μέσω ειδικών μεταγραφικών παραγόντων και ειδικών ρυθμιστών της χρωματίνης. To σύστημα αναφοράς που χρησιμοποιούμε είναι η μεταλλορυθμιζόμενη μεταγραφή στον ζυμομύκητα (Saccharomyces cerevisiae), που αποτελεί κομβικό σημείο στην ομοιοστασία των μετάλλων. Ρυθμιστές της μεταγραφής, που προσδένονται ειδικά στο DNA, μεταβάλλονται λειτουργικά από τη διαθεσιμότητα των μετάλλων, η πρωτεϊνη Mac1 από ιόντα χαλκού και η Aft1 από ιόντα σιδήρου, καθώς και από μεταμεταφραστικές τροποποιήσεις και ειδικές αλληλεπιδράσεις με άλλες πρωτεϊνες.
Η μελέτη δυαδικών και πολυπρωτεϊνικών αλληλεπιδράσεων αποκαλύπτει νέους ρόλους πρωτεϊνών και νέα δεδομένα για μηχανισμούς οργάνωσης της χρωματίνης και της ίδιας της μεταγραφής καθώς και νέες διασυνδέσεις της μεταγραφής (μεταλλορυθμιζόμενης ή μη) με άλλα σηματοδοτικά μονοπάτια (απόκρισης σε βλάβες DNA, οξειδωτικού στρες, κλπ). Ο S. cerevisiae είναι από τα καλύτερα γενετικά χαρακτηρισμένα μοντέλα συστήματα, διαθέτει πλήρη αλληλουχία χαρακτηρισμένου γονιδιώματος, ποικιλία εργαλείων και δοκιμασιών για γενετικές, γονιδιωματικές και πρωτεϊνωματικές αναλύσεις και πλήθος γονιδίων και κυτταρικών διαδικασιών που είναι συντηρημένα ακόμα και στα θηλαστικά.
